You start in Tamayoribito's Cabin. As you leave the cabin, notice that your controller will begin to vibrate. Find the nearby chest and open it to reveal the Essence of Amahagane inside. Using three of these pieces, you can level up your Health, Attack and Defense abilities through the pause screen menu.

Just outside the cabin is Minazuru who can teach about the world of Genji. At the other end of the cabin is a doorway that leads to the cabin's backyard. Do some searching near the well and you'll uncover another Essence of Amahagane. When you've got that, leave the cabin to initiate a Kamui tutorial.
As you leave the tutorial and move away from the cabin, you'll be presented with a world map that shows the areas you can visit. To progress, you'll need to continue to the Main Road area, but don't run there so quickly. Instead, return to Mt. Kurama where you'll find a Broken Sword piece that you can use later. You'll have to fight off more samurai near this pool of water before you can locate the Essence of Amahagane that's hidden nearby. In order to find it, 'follow' the vibration of your controller—as you get closer to the fragment, the vibration will get stronger. Strike the air when you're near the piece to reveal it, then backtrack further to find more Essence of Amahagane.
There are three more Essences of Amahagane to find in the area. One is atop the waterfall, near the area where you located the Iron Sword. The second is located atop one of the platforms near the mid-level savepoint. The third Essence of Amahagane is located at the very beginning of the level, near the building where your journey began. Fight through the samurai that have reinhabited the area to collect the essence, then return to the water to exit the area.
When you return to the world map, head to the Main Road area. There are no enemies to worry about here, though there are merchants you can deal with and some hidden items to find.
Just past the first merchant, look to the north to find a horse stable. At the far end of the stable is a chest you can open to reveal a Kurama Herb. Leave the stable and run around the back side of the building to find another chest and an Essence of Amahagane.
Further up the road, the path branches northward towards a swordsmith shop. Just left of the save point is a small shrine which, upon inspection, gives you another Essence of Amahagane. You can purchase new weapons and armor from the swordsmith if you'd like, or simply head to the west end of the road where a cutscene will stop you and reveal your next mission.
Once back at the world map, head towards Gojo to the south. Upon arrival, you'll be immediately attacked by more Heishi samurai. Defeat them to trigger a cutscene that shows you stumble across the woman you're here to rescue.
Of course, it wouldn't be a good rescue without a dramatic boss fight to kick it off. You'll encounter more samurai headed by Moritoshi Shoki Taira, a powerful wielder of Amahagane. If you've got your Kamui meter built up already, trigger it immediately. Otherwise, wail on the lesser enemies while avoiding Moritoshi to build your meter up so that you can unleash the power of Kamui.

Once you've trigger Kamui, you'll be attacked first by any remaining underlings of Moritoshi before the boss attacks you himself. If you can properly time the attack, you'll bring Moritoshi to his knees and knock off Hoe Goblin's Gauntlet, a piece of armor that can be used for forging later on. You can finish him off with normal combos, or attack more of the samurai to build up another Kamui meter.
When attacking Moritoshi head-on, use quick attacks so that you can interrupt them with an evasive move. When the enemy rears up to attack, hold the guard button and press jump to dodge in the direction you push. Using this move, you can pass right through Moritoshi's attacks and follow up with another quick combo as he recovers from the recoil of his strike.
